M9 medium broth powder, for biotechnology
M9 medium broth powder is a minimal medium, based on the M9 minimal salts, which supports the growth of Escherichia coli. Can maintain bacteria for use with M13 phage.

This medium is useful in maintaining positive selection pressure on plasmids coding for substances, such as amino acids or vitamins. It can also be used to maintain bacteria for use with M13. M9 broth can be supplemented with specific amino acids or other metabolites that allows for specific selection pressures that the researcher may require.

Suspend 10,5 g of M9 medium broth powder in 1 l of distilled, deionised water. Allow the powder to dissolve with gentle stirring and heating, if necessary. Adjust the pH to 7,4 with sodium hydroxide. Autoclave for 15 minutes at 15 psi and at 121 °C.

1 l of media contains 6 g of sodium phosphate dibasic, 3 g of potassium phosphate monobasic, 0,5 g of sodium chloride and 1 g of ammonium chloride.

Store M9 powders at room temperature (18 to 26 °C). Store prepared media cold (2 to 8 °C).

Caution: Note: Alternatively, medium may be sterile-filtered. Allow the solution to cool to 50 °C, or below, before adding supplemental components and antibiotics. Add 2 ml of sterile 1 M magnesium sulphate, 10 ml of sterile 20% glucose and 0,1 ml of sterile 1 M calcium chloride.
